چکیده
The highly diffuse nature of light propagation in biological tissue is a major challenge for obtaining high-fidelity fluorescence tomographic images. In this work we investigated the use of time-gated detection of early-arriving photons for reducing the effects of light scatter in mice relative to quasi-cw photons. When analyzing sinographic representations of the measured data, it was determined that early photons allowed a reduction in the measured FWHM of fluorescent targets by a factor of approximately 2-3, yielding a significant improvement in the tomographic image reconstruction quality.

Tomographic lifetime imaging using combined early- and late-arriving photons.
We present a novel, hybrid approach for time domain fluorescence tomography that efficiently combines lifetime multiplexing using late-arriving or asymptotic photons, with the high spatial resolution capability of early photon tomography. We also show that a decay amplitude-based asymptotic approach is superior to direct inversion of late-arriving photons for tomographic lifetime imaging within...
متن کاملHybrid use of early and quasi-continuous wave photons in time-domain tomographic imaging for improved resolution and quantitative accuracy
Measurement of early-photons (EPs) from a pulsed laser source has been shown to improve imaging resolution versus continuous wave (CW) systems in diffuse optical tomography (DOT) and fluorescence mediated tomography (FMT). However, EP systems also have reduced noise performance versus CW systems since EP measurements require temporal rejection of large numbers of transmitted photons. In this wo...
متن کاملFluorescence tomographic imaging in turbid media using early-arriving photons and Laplace transforms.
We present a multichannel tomographic technique to detect fluorescent objects embedded in thick (6.4 cm) tissue-like turbid media using early-arriving photons. The experiments use picosecond laser pulses and a streak camera with single photon counting capability to provide short time resolution and high signal-to-noise ratio. The tomographic algorithm is based on the Laplace transform of an ana...
متن کاملIn vivo tomographic imaging of deep-seated cancer using fluorescence lifetime contrast.
Preclinical cancer research would benefit from noninvasive imaging methods that allow tracking and visualization of early-stage metastasis in vivo. Although fluorescent proteins revolutionized intravital microscopy, two major challenges that still remain are tissue autofluorescence and hemoglobin absorption, which act to limit intravital optical techniques to large or subcutaneous tumors. Here,...
متن کاملA Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy
Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...
